Characterization of the allergen Sol gem 2 from the fire ant venom, Solenopsis geminata J. Venom. Anim. Toxins incl. Trop. Dis.
Sukprasert,S; Uawonggul,N; Jamjanya,T; Thammasirirak,S; Daduang,J; Daduang,S.
Sol i 2 is a potent allergen in Solenopsis invicta venom, and most humans exhibit reactivity to it. The Sol gem 2 allergen found in the venom of the Thai tropical fire ant Solenopsis geminata was analysed in the present study. The protein was present in higher amounts than other proteins, as determined by SDS-PAGE, and presumably has allergenic properties similar to those of Sol i 2. Sol gem 2 molecular weight is 28 and 15 kDa, respectively, under non-reducing and reducing conditions, indicating that its native form is a dimer. LC-MS/MS analysis confirmed its similarity to Sol i 2. The mono/dimeric form of Sol gem 2 was determined to be relevant by proteomic approach and immunoblotting. An anti-Sol gem 2 antibody was produced in mice, with a titer greater...

Effects of sciatic nerve transection on ultrastructure, NADPH-diaphorase reaction and serotonin-, tyrosine hydroxylase-, c-Fos-, glucose transporter 1- and 3-like immunoreactivities in frog dorsal root ganglion BJMBR
Rigon,F.; Rossato,D.; Auler,V.B.; Dal Bosco,L.; Faccioni-Heuser,M.C.; Partata,W.A..
Frogs have been used as an alternative model to study pain mechanisms. Since we did not find any reports on the effects of sciatic nerve transection (SNT) on the ultrastructure and pattern of metabolic substances in frog dorsal root ganglion (DRG) cells, in the present study, 18 adult male frogs (Rana catesbeiana) were divided into three experimental groups: naive (frogs not subjected to surgical manipulation), sham (frogs in which all surgical procedures to expose the sciatic nerve were used except transection of the nerve), and SNT (frogs in which the sciatic nerve was exposed and transected). After 3 days, the bilateral DRG of the sciatic nerve was collected and used for transmission electron microscopy. Immunohistochemistry was used to detect...

Immunoreactivity for Gonadotropin-Releasing Hormone in Microdialyzed Perfusates of Bovine Mature Follicles In Vitro OAK
Ozawa, Takahiro; Acosta, Tomas J.; Nakamura, Tadashi; Miyamoto, Akio; 中村, 正; 宮本, 明夫.
The presence of gonadotropin-releasing hormone (GnRH)-like protein has been previously demonstrated in the bovine follicle. This GnRH-like protein was purified and concluded to be histone H2A. However, neither GnRH peptide nor specific GnRH-immunoreactivity (GnRH-IR) has been demonstrated in the bovine follicle so far. Thus, this study focused on the detection of specific GnRH-IR using the second-antibody enzyme immunoassay in isolated bovine mature follicles, and on an examination of the direct effect of luteinizing hormone (LH), endothelin-1 (ET-1), and cytokines on the GnRH-IR using an in vitro microdialysis system (MDS). We further examined a cross-reactivity of the GnRH antibody with bovine histone H2A. GnRH-IR was detected in microdialyzed perfusate...

Repeated Binge-Like Ethanol Administration During Adolescence cause Decreased C-Fos Immunoreactivity in Amygdala and Arcuate Nucleus in Adult Sprague-Dawley Rats International Journal of Morphology
Lerma-Cabrera,Jose Manuel; Carvajal,Francisca; Alcaraz-Iborra,Manuel; Sanchez-Amate,Maria del Carmen; de la Fuente,Leticia; Cubero,Inmaculada.
Binge alcohol drinking during adolescence has been associated with neurotoxicity and increased risk for the development of alcohol use disorders. There is evidence that acute and chronic ethanol administration alters c-fos expression, an indirect index of cellular activity, in different brain regions in adult rats. We evaluate here if a binge-like pattern of ethanol exposure during adolescence has a relevant impact on basal and/or ethanol-stimulated regional c-fos activity during adulthood. For that aim, Sprague-Dawley rats PND 25 were saline pre-treated, (SP group) or binge-ethanol pre-treated (BEP group) for twoconsecutive days, at 48-h intervals, over a 14-day period (PND 25 to PND 38). At adult stage (PND 63) and following 25 ethanol-free days, we...

Tryptophan overloading activates brain regions involved with cognition, mood and anxiety Anais da ABC (AABC)
SILVA,LUANA C.A.; VIANA,MILENA B.; ANDRADE,JOSÉ S.; SOUZA,MELYSSA A.; CÉSPEDES,ISABEL C.; D’ALMEIDA,VÂNIA.
ABSTRACT Tryptophan is the only precursor of serotonin and mediates serotonergic activity in the brain. Previous studies have shown that the administration of tryptophan or tryptophan depletion significantly alters cognition, mood and anxiety. Nevertheless, the neurobiological alterations that follow these changes have not yet been fully investigated. The aim of this study was to verify the effects of a tryptophan-enriched diet on immunoreactivity to Fos-protein in the rat brain. Sixteen male Wistar rats were distributed into two groups that either received standard chow diet or a tryptophan-enriched diet for a period of thirty days. On the morning of the 31st day, animals were euthanized and subsequently analyzed for Fos-immunoreactivity (Fos-ir) in the...
